Overview

M6620 + gemcitabine + cisplatin is an investigational **combination regimen** for the treatment of advanced solid tumors and ovarian cancer, among other malignancies. **M6620** (also known as VX-970, berzosertib, or VE-822) is a potent, selective, first-in-class small molecule inhibitor of **ATR kinase** (ataxia telangiectasia and Rad3-related protein), an enzyme involved in cellular DNA damage response. By inhibiting ATR, M6620 blocks cancer cells' ability to repair DNA damage, especially enhancing the cytotoxicity of DNA-damaging chemotherapeutics such as **gemcitabine** (a nucleoside analog antimetabolite) and **cisplatin** (a platinum-based DNA crosslinker). This combination has been studied in platinum-resistant and refractory ovarian cancer as well as other advanced solid tumors, showing evidence of increased DNA damage and potential clinical activity when ATR inhibition is added to cytotoxic chemotherapy[2][5].
