Overview

M72 + AS01B is an investigational **recombinant subunit vaccine candidate for tuberculosis (TB)**, composed of the M72 antigen and the AS01B adjuvant system. The **M72 antigen** is a recombinant fusion protein derived from two immunogenic *Mycobacterium tuberculosis* proteins, Mtb32A and Mtb39A, chosen for their ability to stimulate strong T-cell immune responses. The **AS01B adjuvant** is a liposome-based system containing two immunostimulants: monophosphoryl lipid A (MPL), a TLR4 agonist, and saponin QS-21 from Quillaja saponaria, designed to enhance both humoral and cellular immune responses—particularly robust CD4+ T-cell (Th1) responses and antibody production[1][2]. The vaccine is delivered intramuscularly and is currently in late-stage (PHASE3) clinical development for the prevention of active tuberculosis in infected or at-risk adults.
