Mab0727 (MMAE) is an investigational **CDH17-targeted antibody-drug conjugate** comprising the humanized anti-cadherin-17 monoclonal antibody Mab0727 conjugated to the microtubule-disrupting cytotoxic payload monomethyl auristatin E. Binding of the antibody to CDH17 on gastrointestinal cancer cells promotes internalization of the conjugate, enabling intracellular release of MMAE and inhibition of tubulin polymerization, causing mitotic arrest and tumor-cell death. The construct was used as a preclinical MMAE-based comparator in Mabwell's CDH17 ADC research and had a drug-to-antibody ratio of approximately 4. It is distinct from 7MW4911, which uses the same Mab0727 antibody but a different MF-6 topoisomerase I inhibitor payload.
