Overview

This combination therapy consists of macitentan and dapagliflozin and is being investigated by Gachon University Gil Medical Center for the treatment of combined pre- and post-capillary pulmonary hypertension (CpcPH) in patients with heart failure with mildly reduced ejection fraction (HFmrEF) and heart failure with preserved ejection fraction (HFpEF). Macitentan is a dual endothelin receptor antagonist (ERA) that blocks both endothelin A (ET-A) and endothelin B (ET-B) receptors, which helps to reduce pulmonary vascular resistance and improve hemodynamics. Dapagliflozin is a sodium-glucose co-transporter 2 (SGLT2) inhibitor that reduces glucose reabsorption in the kidneys and has established cardiovascular benefits in heart failure management. The combination is currently being evaluated in a Phase 4 exploratory clinical trial to assess its efficacy and safety in this specific patient population.
