MAG-TN3 + AS15 is a fully synthetic glycopeptide cancer vaccine designed to induce an immune response against the Tn antigen, a tumor-associated carbohydrate antigen expressed on several adenocarcinomas but not on normal cells. The vaccine consists of the multiple antigenic glycopeptide (MAG-Tn3), which displays three Tn moieties and includes a tetanus toxoid-derived CD4+ T-cell epitope (TT830–844) to enhance helper T-cell responses. It is formulated with the proprietary immunostimulant AS15, which contains CpG7909 (a Toll-like receptor 9 agonist), monophosphoryl lipid A (MPL, a Toll-like receptor 4 agonist), and QS21. Upon administration, MAG-TN3 induces production of tumor-specific anti-Tn antibodies that can mediate complement-dependent cytotoxicity against Tn-expressing tumor cells. The combination has been shown in phase I clinical trials to be well tolerated and highly immunogenic in patients with high-risk localized breast cancer[1][2][3][4][5].
