A combination therapy consisting of MAGE-A3 immunotherapeutic (also known as astuprotimut-R or recMAGE-A3+AS15, a cancer vaccine containing recombinant MAGE-A3 protein and AS15 immunostimulant) with cisplatin and vinorelbine chemotherapy. The MAGE-A3 component is designed to target MAGE-A3, a tumor-specific antigen expressed in various cancers including melanoma, non-small cell lung cancer (NSCLC), and head and neck cancer, but not in normal cells. The AS15 immunostimulant contains 3-O-desacyl-4′-monophosphoryl lipid A (MPL), Quillaja saponaria Molina fraction 21 (QS-21), and CpG 7909 synthetic oligodeoxynucleotides in a liposomal formulation, designed to enhance the immune response. In preclinical studies, the MAGE-A3 immunotherapeutic showed promising results, inducing antigen-specific, functional, and long-lasting immune responses capable of recognizing and eliminating MAGE-A3-expressing tumor cells. Its mechanism of action involves stimulating both humoral and cellular immune responses against MAGE-A3-expressing tumor cells.
