MAGE-A4-specific TCR-T cells (Sun Yat-Sen Memorial Hospital) is an autologous T-cell receptor (TCR) engineered T-cell therapy designed to target the Melanoma-associated antigen A4 (MAGE-A4). MAGE-A4 is a cancer-testis antigen that is frequently overexpressed in various solid tumors, particularly mesenchymal malignancies such as synovial sarcoma and myxoid/round cell liposarcoma, while its expression in normal tissues is restricted to the testis and placenta. The therapy involves harvesting a patient's T cells, genetically engineering them to express a TCR specific for a MAGE-A4 peptide presented by HLA-A*02:01, and expanding them ex vivo before re-infusion. This approach leverages the patient's immune system to specifically recognize and eliminate MAGE-A4-positive cancer cells. It is currently being investigated in a Phase 1 clinical trial for patients with advanced mesenchymal malignancies.
