Overview

Magnesium sulfate + sodium bicarbonate is an investigational combination therapy developed by researchers at West China Hospital, Sichuan University, for the treatment of knee osteoarthritis. Administered via intra-articular injection, the formulation utilizes a dual mechanism to address joint pain and cartilage degradation. Magnesium sulfate acts as a non-competitive N-methyl-D-aspartate (NMDA) receptor antagonist and a calcium channel blocker, providing analgesic and anti-inflammatory effects by inhibiting pain signaling pathways. Sodium bicarbonate serves as an alkalizing agent that neutralizes the acidic microenvironment (acidosis) typically found in osteoarthritic joints, which is associated with increased pain sensitization and accelerated cartilage breakdown. By combining these agents, the therapy aims to improve joint function and provide symptomatic relief by modifying the biochemical and physical environment of the synovial space.
