magrolimab + bevacizumab + FOLFIRI

Magrolimab + bevacizumab + FOLFIRI is an investigational combination therapy developed by Gilead Sciences for the treatment of previously treated advanced inoperable metastatic colorectal cancer (mCRC). Magrolimab is a first-in-class monoclonal antibody targeting CD47, an immune checkpoint that cancer cells use to avoid being eaten by macrophages (the "don't eat me" signal). Bevacizumab is a vascular endothelial growth factor (VEGF) inhibitor that inhibits angiogenesis. FOLFIRI is a standard chemotherapy regimen consisting of folinic acid (leucovorin), 5-fluorouracil, and irinotecan. The combination is designed to leverage the synergistic effects of CD47 blockade, anti-angiogenesis, and chemotherapy-induced prophagocytic signals to enhance antitumor activity. It is currently being evaluated in the Phase 2 ELEVATE CRC trial (GS-US-587-6156).
