Overview

**Magrolimab + CC-486** is a combination investigational regimen composed of magrolimab (a monoclonal antibody targeting cluster of differentiation 47, CD47) and CC-486 (the oral formulation of azacitidine, marketed as ONUREG), evaluated primarily in myeloid malignancies including acute myeloid leukemia (AML). Magrolimab inhibits the "don't eat me" signal mediated by CD47 on malignant cells, promoting phagocytosis by macrophages. CC-486 (oral azacitidine) is a DNA methyltransferase inhibitor that induces hypomethylation of DNA, thereby restoring normal gene function and promoting differentiation or apoptosis of malignant cells. The combination is under clinical investigation for potential synergy, given CC-486's ability to upregulate "eat-me" signals, possibly enhancing magrolimab's antitumor effect[3][5].
