Overview

Magrolimab + mitoxantrone + etoposide + cytarabine is a combination therapy studied specifically for the treatment of relapsed/refractory acute myeloid leukemia (AML). Magrolimab is a monoclonal antibody targeting CD47, a "don't eat me" signal that helps cancer cells evade phagocytosis, thus enabling immune-mediated elimination of malignant myeloid cells. Mitoxantrone, etoposide, and cytarabine (collectively known as MEC) are cytotoxic chemotherapeutic agents commonly used in salvage regimens for relapsed/refractory AML. Magrolimab is hypothesized to enhance the effector function of chemotherapy by augmenting immune cell-mediated clearance of leukemia cells, possibly through the upregulation of prophagocytic signals such as calreticulin after chemotherapy-induced cell stress. This regimen is investigational and not approved for marketing, but a Phase 2 multi-arm study (NCT04778410) demonstrated its feasibility and manageable safety profile in patients with relapsed/refractory AML[3][5][6][7].
