Drug intelligence / Profile preview
This combination consists of **magrolimab**, a monoclonal antibody targeting CD47; **pembrolizumab**, a monoclonal antibody targeting programmed cell death protein 1 (PD-1); **5-fluorouracil (5-FU)**, a pyrimidine analog and antimetabolite chemotherapeutic; and **platinum chemotherapy**, which typically refers to cisplatin or carboplatin, platinum-based DNA crosslinking chemotherapeutic agents. - Magrolimab blocks the CD47 "don't eat me" signal, promoting macrophage-mediated phagocytosis of tumor cells. - Pembrolizumab inhibits PD-1, lifting suppression on antitumor T-cell responses to enhance immune-mediated tumor cell killing. - 5-fluorouracil inhibits thymidylate synthase, disrupting DNA synthesis and promoting cell death in proliferating cancer cells. - Platinum chemotherapies form DNA adducts and crosslinks, inducing apoptosis primarily in rapidly proliferating cells. This combination is under phase 2 investigation for the treatment of **untreated metastatic or unresectable, locally recurrent head and neck squamous cell carcinoma (HNSCC)**[4][5].
