Manganese(II)-N,N'-bis(salicylidene)-1,2-phenylenediamine is a manganese Schiff base (salen-type) complex developed by Hebei University of Science and Technology and Hebei Medical University for the potential treatment of type 2 diabetes. It functions as a noncompetitive inhibitor of dipeptidyl peptidase IV (DPP-IV), an enzyme responsible for the degradation of incretin hormones like GLP-1. In preclinical research, the compound exhibited moderate inhibitory activity against DPP-IV with an IC50 of 30 μM and a Ki of 136.3 μM. In vivo testing in alloxan-induced diabetic mice demonstrated that oral (intragastric) administration of the complex significantly lowers serum glucose levels. Molecular modeling suggests the complex binds within the active-site cleft of the DPP-IV enzyme. Furthermore, the compound showed a favorable safety profile in acute toxicity studies with no significant abnormal signs observed at high doses.
