Overview

manganese dioxide + ovalbumin + A50-T20 is an experimental nanoparticle-based tumor vaccine designed to optimize lymph node delivery and antigen-presenting cell (APC) uptake. The formulation addresses the "granularity paradox"—where small particles are needed for efficient drainage to lymph nodes, but larger particles are better for APC capture—by using a DNA-mediated aggregation strategy. It consists of hollow mesoporous manganese dioxide (MnO2) nanoparticles loaded with the model antigen ovalbumin (OVA) and functionalized with complementary DNA single strands (A50 or T20). When administered sequentially, the small (~90 nm) particles drain to the lymph nodes and aggregate into larger clusters via DNA base-pairing. This in situ enlargement enhances antigen delivery and stimulates a robust anti-tumor cellular immune response. The MnO2 component also acts as an adjuvant and can modulate the tumor microenvironment by alleviating hypoxia.
