maraviroc + dapivirine

Maraviroc + dapivirine is a combination of two antiretroviral agents being investigated primarily as a topical microbicide for the prevention of sexual transmission of HIV. Maraviroc is a small molecule CCR5 antagonist that blocks HIV entry into human cells by preventing the interaction between the virus and the chemokine receptor type 5 (CCR5) co-receptor on CD4+ T-cells. Dapivirine is a non-nucleoside reverse transcriptase inhibitor (NNRTI) that inhibits HIV-1 reverse transcriptase, thereby blocking viral replication. The combination has been formulated in vaginal rings and gels to provide localized, sustained drug delivery for pre-exposure prophylaxis (PrEP). Clinical studies have shown that both drugs are generally safe and well-tolerated when used together in vaginal rings, though dapivirine achieves higher tissue concentrations than maraviroc in this formulation[2][4][5]. The primary indication under investigation is HIV prevention.
