MART-1:27-35(27L) is a synthetic heteroclitic peptide vaccine derived from the melanoma-associated antigen MART-1 (also known as Melan-A). This specific peptide represents a nine-amino acid sequence corresponding to positions 27 through 35 of the native MART-1 protein, but with a strategic substitution of Alanine to Leucine at position 27 (LAGIGILTV). This modification enhances the peptide's binding affinity for the HLA-A*0201 MHC class I molecule, acting as an 'anchor residue' substitution that increases the stability of the peptide-MHC complex. Upon subcutaneous administration, typically in conjunction with an adjuvant like Montanide ISA-51, the peptide is presented by antigen-presenting cells to CD8+ T cells. This process stimulates the expansion of MART-1-specific cytotoxic T lymphocytes (CTLs), which are then capable of recognizing and lysing melanoma cells that express the native MART-1 antigen. Developed largely by the National Cancer Institute (NCI), this peptide has been investigated in various Phase 1 and Phase 2 clinical trials as an immunotherapy for patients with metastatic melanoma.
