This investigational multi-epitope peptide vaccine was developed by Ahmad Tarhini and colleagues at the University of Pittsburgh Cancer Institute for the treatment of advanced (Stage III/IV) melanoma. The vaccine formulation includes three melanoma-associated peptide antigens: MART-1 (26-35, 27L), gp100 (209-217, 210M), and tyrosinase (368-376, 370D). To maximize immunogenicity, these peptides are combined with two potent adjuvants: sargramostim (GM-CSF), which promotes dendritic cell maturation, and agatolimod (PF-3512676), a Toll-like receptor 9 (TLR9) agonist that stimulates innate and adaptive immune responses. The entire mixture is emulsified in Montanide ISA-51, an incomplete Freund's-like adjuvant. The vaccine is designed to elicit specific CD8+ T-cell responses against melanoma cells in HLA-A2+ patients.
