Drug intelligence / Profile preview
Matrix-M2 is a proprietary saponin-based vaccine adjuvant developed by Novavax (formerly Isconova) and licensed for use in various clinical programs, most notably Genocea Biosciences' GEN-003 therapeutic vaccine for genital herpes (HSV-2). It is composed of purified saponin fractions from the *Quillaja saponaria* Molina tree, which are formulated with cholesterol and phospholipids into cage-like nanoparticles approximately 40 nm in size, known as Immune Stimulating Complexes (ISCOMs). Matrix-M2 functions by enhancing the recruitment and activation of antigen-presenting cells at the injection site and in draining lymph nodes, thereby promoting a balanced and potent immune response characterized by high antibody titers and robust CD4+ and CD8+ T-cell activity. Although the GEN-003 program was terminated following Genocea's closure, the Matrix-M adjuvant technology remains a core component of Novavax's portfolio, including their authorized COVID-19 and malaria vaccines.
