This is a **combination therapy** comprising four drugs: **matuzumab**, a humanized IgG1 monoclonal antibody targeting epidermal growth factor receptor (**EGFR**), with **epirubicin** (an anthracycline topoisomerase II inhibitor), **cisplatin** (a platinum-based DNA crosslinker), and **capecitabine** (a prodrug of 5-fluorouracil). It was studied as first-line treatment for advanced, EGFR-positive oesophagogastric adenocarcinoma. Matuzumab blocks ligands from binding to EGFR, thereby inhibiting receptor-mediated downstream signaling and tumor cell proliferation; it can also trigger antibody-dependent cytotoxicity. The other three components are established chemotherapy agents with cytotoxic antitumor properties.
