Mavrostobart + pemetrexed is a combination of two investigational anticancer agents being evaluated primarily for solid tumors including non-small cell lung cancer (NSCLC) and potentially other malignancies. Mavrostobart is a monoclonal antibody that targets and inhibits CD73, an ectoenzyme on tumor and stromal cells involved in generating adenosine, which contributes to an immunosuppressive tumor microenvironment. Mavrostobart is designed to fully inhibit both soluble and membrane-bound CD73, thereby counteracting adenosine-mediated immune suppression and enhancing antitumor immunity. Pemetrexed is a small molecule antimetabolite and multitargeted antifolate that inhibits several folate-dependent enzymes essential for nucleotide synthesis and tumor cell proliferation. Pemetrexed is widely used in combination chemotherapy in NSCLC and mesothelioma. The combination is currently being evaluated in a multicenter Phase I/II clinical trial (MORNINGSTAR study) to assess safety, tolerability, pharmacokinetics, and preliminary efficacy in patients with advanced cancer, with a specific focus on its potential to improve immune response in the tumor microenvironment[2][4].
