Overview

MAX-40279-01 + toripalimab is an experimental **combination therapy** consisting of **MAX-40279-01**, a novel multi-target tyrosine kinase inhibitor, and **toripalimab**, an anti-PD-1 monoclonal antibody. MAX-40279-01 directly inhibits multiple receptor tyrosine kinases (notably FGFR, FLT3, and also JAK, HPK1, PDGFRs, VEGFR), and suppresses hematopoietic progenitor kinase 1 (HPK1) to relieve negative regulation of the T cell receptor (TCR) pathway, thereby enhancing anti-tumor immune activity. Toripalimab works by blocking the PD-1 receptor on T cells, preventing its interaction with PD-L1/PD-L2 on tumor cells, which facilitates immune system recognition and attack of cancer cells. This combination is being jointly developed to target advanced solid tumors, leveraging dual mechanisms: targeted kinase inhibition and immune checkpoint blockade[1][3][7]. MAX-40279-01 was developed by Maxinovel as its first NME and is being evaluated in combination with toripalimab in multicenter Phase I studies for advanced solid tumors[1]. Toripalimab was developed by Junshi Biosciences and is already approved in China for various cancers[1]. The combination aims to overcome resistance to immunotherapy and expand the clinical benefit in solid tumors with high immune tolerance.
