A three-drug combination regimen consisting of **MBP-426** (a transferrin-conjugated liposomal formulation of oxaliplatin), **leucovorin** (folinic acid), and **5-fluorouracil** used in the treatment of advanced or metastatic solid tumors, particularly for gastrointestinal cancers such as gastric, gastro-esophageal, or esophageal adenocarcinoma. MBP-426 is designed for enhanced tumor targeting via transferrin receptor-mediated uptake and the enhanced permeability and retention effect, potentially reducing neurotoxicity seen with standard oxaliplatin. Its mechanism involves DNA crosslinking by oxaliplatin, while 5-fluorouracil inhibits thymidylate synthase and leucovorin potentiates 5-FU cytotoxicity[2][7][8].
