Overview

MBP431 is an investigational pan-ebolavirus monoclonal antibody cocktail developed by Mapp Biopharmaceutical. It is a second-generation, half-life extended version of MBP134, consisting of two broadly neutralizing human monoclonal antibodies, ADI-15878YTE and ADI-23774YTE. These antibodies target highly conserved, non-overlapping epitopes on the ebolavirus glycoprotein (GP) and have been engineered with YTE mutations (M252Y/S254T/T256E) in their Fc regions to prolong serum half-life and enable intramuscular (IM) administration. MBP431 is designed for post-exposure prophylaxis (PEP) and treatment of Ebola virus disease (EVD) caused by multiple ebolavirus species, including Zaire ebolavirus (EBOV), Sudan ebolavirus (SUDV), and Bundibugyo ebolavirus (BDBV). Preclinical studies in non-human primates have demonstrated that a single IM dose of MBP431 provides significant protection against lethal ebolavirus challenges, and its combination with the antiviral remdesivir further enhances survival outcomes in advanced stages of disease.
