MBS8 (1V270) is a Toll-like receptor 7 (TLR7) agonist developed by MonTa Biosciences, formulated as a phospholipid-conjugated small molecule within micelles for intravenous administration. This micellar formulation is designed to enable systemic delivery while minimizing the toxicities typically associated with TLR7 activation, which are usually restricted to topical use. Keytruda (pembrolizumab) is a humanized monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor, functioning as an immune checkpoint inhibitor. The combination of MBS8 and pembrolizumab aims to synergistically activate both innate and adaptive immune responses to treat advanced solid tumors, particularly in cases where tumors have become resistant to anti-PD-1 monotherapy. Clinical evaluation is currently focused on advanced solid tumors, including cutaneous and uveal melanoma, through the Phase I MBS8-101 trial.
