Overview

MCLA-129 + chemotherapy is a combination regimen consisting of **MCLA-129**, a bispecific monoclonal antibody targeting both Epidermal Growth Factor Receptor (**EGFR**) and hepatocyte growth factor receptor (**c-MET**), together with cytotoxic chemotherapy agents (not specified, generally platinum-based chemotherapy in non-small cell lung cancer contexts). MCLA-129 is engineered for enhanced antibody-dependent cellular cytotoxicity and antibody-dependent cellular phagocytosis, and works by inhibiting ligand-dependent phosphorylation of EGFR and c-MET as well as overcoming resistance to tyrosine kinase inhibitors. The combination aims to increase antitumor activity in patients with advanced or metastatic solid tumors, such as non-small cell lung cancer (NSCLC) harboring MET exon 14 skipping mutation (METex14), EGFR exon20 insertion (exon20ins), and sensitizing EGFR mutations. In clinical studies, patients previously treated with platinum-based chemotherapy or tyrosine kinase inhibitors were included[1][5].
