MCSP:TRAIL is a recombinant fusion protein designed for the targeted treatment of melanoma. It consists of a single-chain variable fragment (scFv) specific for the Melanoma-associated Chondroitin Sulfate Proteoglycan (MCSP), also known as Chondroitin Sulfate Proteoglycan 4 (CSPG4), genetically fused to the soluble extracellular domain of the tumor necrosis factor-related apoptosis-inducing ligand (TRAIL). By binding to MCSP, which is highly expressed on the surface of melanoma cells, the fusion protein concentrates TRAIL at the tumor site. This localized delivery facilitates the trimerization and activation of TRAIL receptors (DR4 and DR5), thereby inducing the extrinsic apoptotic pathway specifically in cancer cells while sparing normal tissues. Preclinical studies have demonstrated its efficacy in inhibiting melanoma growth both in vitro and in vivo.
