MDNA11 + pembrolizumab is an investigational combination regimen comprising two agents: MDNA11, a long-acting, "beta-only" recombinant interleukin-2 (IL-2)-albumin fusion protein, and pembrolizumab (Keytruda), a monoclonal antibody immune checkpoint inhibitor targeting PD-1. MDNA11 is engineered to enhance stimulation of effector immune cells (CD8+ T-cells and NK cells) while avoiding stimulation of regulatory T-cells, thereby intensifying the anti-tumor immune response. Pembrolizumab acts by blocking PD-1-mediated immune inhibition, reactivating T-cell responses against tumor cells. This combination is in clinical trials for the treatment of various advanced solid tumors, including but not limited to clear cell renal cell carcinoma, resistant/refractory melanoma, and MSI-H pancreatic cancer, with the intention of leveraging synergistic immunomodulation to improve anti-tumor efficacy[1][2][3][4][5].
