Overview

MDNA19 + MDNA413 is an investigational **dual-cytokine biologic immunotherapy** from Medicenna that combines the IL-2 super-agonist MDNA19 with the IL-13 super-antagonist MDNA413 in a single BiSKITs-derived construct. The molecule is designed to both stimulate anti-tumor immunity and suppress pro-tumoral type 2 immune signaling: MDNA19 selectively signals through the intermediate-affinity interleukin-2 receptor complex composed of CD122 and CD132 to drive a pro-inflammatory anti-cancer response, while MDNA413 blocks IL-4 and IL-13 signaling through the type 2 IL-4 receptor complex containing IL-4R and IL-13R1. The program has been described as a DUal CytoKine for cancer and has been positioned for treatment of immunologically cold tumors, with preclinical data reported by 2021.
