A **combination therapy** of ME-344, a mitochondrial inhibitor, and topotecan, a topoisomerase I inhibitor, studied primarily for solid tumors including **small cell lung cancer**, **ovarian cancer**, and **cervical cancer**. ME-344 disrupts mitochondrial function and induces cancer cell death, acting independently of caspase-mediated pathways and showing effectiveness against chemotherapy-resistant cancer stem cells in preclinical models. Topotecan inhibits DNA topoisomerase I, leading to DNA damage in S-phase cells. The combination was tested for synergistic antitumor effects but, while tolerable, did not demonstrate sufficient efficacy to warrant further study in unselected advanced ovarian, cervical, or small cell lung cancer populations[1][5][3][4].
