Overview

MEDI-517 + AS04 is a **combination prophylactic vaccine candidate** composed of **MEDI-517**, which is a human papillomavirus (HPV)-16/18 virus-like particle (VLP) vaccine, and **AS04**, a proprietary adjuvant system. The vaccine is designed to induce strong immunity against HPV-16 and HPV-18, which are high-risk serotypes associated with cervical cancer. AS04 contains monophosphoryl lipid A (MPL), a detoxified derivative of lipopolysaccharide that acts as a Toll-like receptor 4 (TLR4) agonist, adsorbed onto aluminum hydroxide (alum). The combination of VLPs with AS04 enhances both humoral and cellular immune responses, inducing higher antibody levels and robust memory B-cell formation versus equivalent vaccines using aluminum only. MEDI-517 was developed and studied as GSK 580299 in collaboration with GSK, with immunogenicity and safety trials in women aged 15–55 years[3][5][1].
