MEDI-551 + bendamustine is a combination therapy composed of MEDI-551, an afucosylated humanized monoclonal antibody targeting CD19, and bendamustine, a cytotoxic alkylating agent. MEDI-551 acts by binding to CD19, a B-cell surface antigen, resulting in antibody-dependent cellular cytotoxicity (ADCC) and B cell depletion. Bendamustine interferes with DNA replication and repair through alkylation, leading to cell death. The combination has been evaluated in phase 2 trials for adults with relapsed or refractory chronic lymphocytic leukemia (CLL). Compared to rituximab plus bendamustine, this combination demonstrated a distinct safety profile and evidence of clinical activity, particularly at higher MEDI-551 doses[2][5][7].
