Overview

MEDI0562 + tremelimumab is an investigational combination therapy composed of two monoclonal antibodies: **MEDI0562**, a humanized IgG1K monoclonal antibody that acts as an agonist of the OX40 receptor (TNFRSF4) on activated T cells, and **tremelimumab**, a human monoclonal antibody that targets cytotoxic T-lymphocyte-associated protein 4 (CTLA-4). This combination is designed to enhance anti-tumor immune responses via dual checkpoint modulation: MEDI0562 stimulates T-cell activation and proliferation by activating OX40 (costimulatory molecule), while tremelimumab blocks CTLA-4, thereby releasing another inhibitory brake on T cells. The therapy is under investigation for the treatment of advanced solid tumors, including ovarian cancer and head and neck squamous cell carcinoma. MEDI0562 is being developed primarily by MedImmune/AstraZeneca[1][2][3][5][9].
