Overview

This is a combination of three agents: **MEDI3617**, **carboplatin**, and **paclitaxel**. - **MEDI3617** is a human monoclonal antibody that selectively inhibits angiopoietin-2 (Ang2), blocking its interaction with the Tie2 receptor and thereby interfering with tumor angiogenesis. - **Carboplatin** is a platinum-based chemotherapeutic agent that induces DNA crosslinks, leading to apoptosis of rapidly dividing cells. - **Paclitaxel** is a microtubule-stabilizing chemotherapeutic agent that disrupts cell division, leading to cell death. This combination was investigated in phase 1 clinical trials for advanced solid tumors and, specifically, platinum-resistant ovarian cancer. MEDI3617 demonstrated acceptable safety in combination with chemotherapy including carboplatin and paclitaxel, and preclinical models supported enhanced antitumor responses compared to monotherapy. However, clinical development of MEDI3617 was discontinued due to limited activity and safety issues, particularly edema in ovarian cancer[1][2][7].
