Overview

MEDI6469 + tremelimumab is an investigational combination of two immunotherapy agents. Tremelimumab is a humanized IgG2 monoclonal antibody that acts as an immune checkpoint inhibitor by binding to cytotoxic T-lymphocyte-associated antigen 4 (CTLA-4), blocking its interaction with ligands CD80 and CD86. This blockade enhances T-cell activation and proliferation, resulting in increased antitumor immune activity[5]. Tremelimumab is developed primarily for the treatment of unresectable hepatocellular carcinoma (HCC) and non-small cell lung cancer (NSCLC), often in combination with other agents such as durvalumab[1][3][8]. The code name MEDI6469 refers to an anti-OX40 monoclonal antibody developed by AstraZeneca/MedImmune for cancer immunotherapy; it acts as an agonist of the OX40 receptor on T cells, further stimulating immune responses against tumors. The combination aims to synergistically enhance antitumor immunity through dual checkpoint modulation.
