Drug intelligence / Profile preview
Megace ES is an advanced oral suspension formulation of megestrol acetate, a synthetic derivative of the naturally occurring steroid hormone progesterone. Developed by Par Pharmaceutical using NanoCrystal technology, Megace ES provides significantly improved bioavailability compared to the original Megace formulation, allowing for a lower dosing volume (625 mg in 5 mL versus 800 mg in 20 mL). It is primarily indicated for the treatment of anorexia, cachexia, or unexplained significant weight loss in patients with acquired immunodeficiency syndrome (AIDS). Megestrol acetate acts as an agonist at the progesterone receptor; its orexigenic effects are thought to be mediated through the ventromedial nucleus of the hypothalamus and the inhibition of pro-inflammatory catabolic cytokines like tumor necrosis factor-alpha (TNF-α), interleukin-1, and interleukin-6, which contribute to metabolic wasting. While megestrol acetate is also used in the treatment of breast and endometrial cancers, the Megace ES formulation is specifically marketed for AIDS-related wasting.
