Megestrol acetate + medroxyprogesterone acetate is a combination of two synthetic progestins used individually in the treatment of various conditions such as advanced breast cancer, endometrial cancer, abnormal uterine bleeding, amenorrhea, and as appetite stimulants in cachexia and weight loss. Both compounds act primarily as agonists of the progesterone receptor, exerting progestogenic effects that can alter hormone-dependent cancer growth and regulate menstrual function. Megestrol acetate also displays weak glucocorticoid and androgenic activity, while medroxyprogesterone acetate is used in higher doses for palliative cancer treatment and as a contraceptive. Combination therapy may theoretically increase progestogenic effects or side effects, with overlapping adverse event profiles such as appetite changes, fluid retention, thromboembolic events, and Cushingoid symptoms. The combination may be considered in specific cases where enhanced progestin effect is desired, such as for certain cancers, but both are usually used as individual agents rather than as a standard combination therapy.[1][2][5][6][7][9]
