Overview

Melphalan + cyclophosphamide is a combination chemotherapy regimen consisting of two alkylating agents, melphalan and cyclophosphamide. Both drugs function by cross-linking DNA strands, thereby inhibiting DNA and RNA synthesis and leading to cell death in both dividing and non-dividing tumor cells. This combination is used as high-dose myeloablative chemotherapy, particularly in the treatment of relapsed or refractory brain tumors (such as medulloblastoma and germinoma) and has also been studied for use in multiple myeloma as part of conditioning regimens prior to autologous stem cell transplantation. The regimen has demonstrated tolerability with notable complete response rates in pediatric oncology settings[1][2][4].
