melphalan + dexamethasone is a combination therapeutic being developed by 3T Ophthalmics for the treatment of intraocular cancers, specifically retinoblastoma and uveal melanoma. The combination consists of melphalan, a potent DNA-alkylating cytotoxic agent, and dexamethasone, a glucocorticoid receptor agonist that provides anti-inflammatory effects. The treatment is delivered using 3T Ophthalmics' proprietary Targeted Episcleral Delivery System (TEDS), a periocular reservoir device placed under the conjunctiva in contact with the episclera. This delivery method enables sustained and controlled release of the chemotherapeutic agents directly to the posterior segment of the eye, achieving high local concentrations while minimizing systemic exposure and associated side effects.
