This drug is a combination chemotherapy regimen consisting of three small molecule agents—melphalan, methotrexate, and mitomycin C. Each component has a distinct mechanism of action: - Melphalan is an alkylating agent that crosslinks DNA strands, inhibiting DNA and RNA synthesis. - Methotrexate is an antimetabolite that inhibits dihydrofolate reductase, blocking folic acid metabolism and thus DNA synthesis. - Mitomycin C acts as a bioreductive alkylating agent causing DNA crosslinking and strand breakage. The combination (sometimes referred to as "MMM") has been studied primarily for metastatic breast carcinoma. Clinical studies have shown it to be effective and generally well tolerated in this setting, with response rates around 40% in previously untreated patients[1]. The regimen may also help control hypercalcemia associated with bone metastases[1]. Cumulative marrow toxicity can occur but was not significant within the first six months of treatment[1].
