Overview

Melphalan + mitoxantrone is a combination chemotherapy regimen consisting of two small molecule antineoplastic agents. Melphalan is an alkylating agent that crosslinks DNA and inhibits DNA and RNA synthesis, leading to cell death. Mitoxantrone is an anthracenedione antineoplastic agent that intercalates into DNA, causing crosslinks and strand breaks, as well as inhibiting topoisomerase II. This combination has been used primarily as a conditioning regimen prior to autologous stem cell transplantation in patients with lymphoma or multiple myeloma[2][4][6][7]. It has also been studied in other hematologic malignancies such as acute myeloid leukemia (AML) and high-risk myelodysplastic syndrome (MDS)[1][5]. The regimen aims to maximize cytotoxicity against malignant cells while allowing for subsequent hematopoietic recovery via stem cell support.
