Overview

MEN1611 + cetuximab is an experimental combination therapy consisting of MEN1611, a potent, orally available, δ-sparing class I PI3K inhibitor, and cetuximab, a chimeric monoclonal antibody targeting the epidermal growth factor receptor (EGFR). MEN1611 selectively inhibits the PI3K catalytic subunits p110α, p110β, and p110γ, with minimal activity on p110δ, aiming to inhibit tumor cell proliferation and survival by blocking the PI3K pathway, particularly in cancers with PIK3CA mutations or PTEN loss. Cetuximab blocks EGFR signaling, which is often upregulated in several cancers, thereby inhibiting downstream proliferative pathways. The combination is under investigation for treating metastatic colorectal cancer, based on synergistic potential to block tumor growth by disrupting both EGFR and PI3K/AKT signaling axes[5].
