MesoPher + mitazalimab is an investigational combination therapy comprising two components: - **MesoPher** is an autologous dendritic cell (DC) vaccine loaded with PheraLys, an allogeneic mesothelioma tumor cell lysate. Dendritic cells are generated ex vivo from a patient's monocytes collected via leukapheresis, loaded with PheraLys (containing tumor-associated antigens), matured, and then administered back to the patient as a personalized immunotherapy to induce a tumor-specific immune response[1][3][5]. - **Mitazalimab** is a monoclonal antibody targeting CD40, a receptor on antigen-presenting cells (primarily dendritic cells, B cells, and macrophages). As a CD40 agonist, it activates dendritic cells, enhancing their ability to stimulate T cells and augmenting antitumor immune responses[2][4][5]. The combination seeks to harness and boost the patient’s immune response by both expanding tumor antigen presentation (via MesoPher) and amplifying immune cell activation (via mitazalimab). It is being tested in adults with metastatic pancreatic cancer, particularly after standard-of-care (mFOLFIRINOX) chemotherapy[5].
