Overview

Metamizole (dipyrone) + tramadol is an analgesic combination consisting of the non-opioid pyrazolone derivative metamizole and the synthetic opioid tramadol. Metamizole provides analgesic, antipyretic, and antispasmodic effects, likely through the inhibition of cyclooxygenase enzymes and modulation of the endocannabinoid system and descending inhibitory pain pathways. Tramadol exerts its effect via weak agonism of the mu-opioid receptor and by inhibiting the reuptake of norepinephrine and serotonin. This specific combination was evaluated in clinical trials (such as NCT01227200) by HaEmek Medical Center as an adjunct to local anesthesia for pain management during the incision and drainage of peritonsillar abscesses (PTA). It is typically used as a co-administration of generic components rather than a proprietary fixed-dose branded product in this research context.
