Overview

**Metformin** is an orally administered small-molecule biguanide antihyperglycemic drug approved for type 2 diabetes mellitus. It lowers glucose principally by reducing hepatic glucose production, decreasing intestinal glucose absorption, and improving peripheral insulin sensitivity. In the University of Cincinnati Phase I study NCT02325401, metformin was repurposed as an investigational adjunct to concurrent cisplatin and radiation therapy for locally advanced head and neck squamous cell carcinoma.
