This treatment regimen involves the use of metformin as an adjuvant to standard chemotherapy protocols for the treatment of metastatic breast cancer. Metformin, a biguanide typically used for type 2 diabetes, is being investigated for its potential to lower insulin-like growth factor 1 (IGF-1) levels, which are often elevated in breast cancer and associated with poor prognosis. By inhibiting the mitochondrial respiratory chain (Complex I) and activating AMP-activated protein kinase (AMPK), metformin may reduce circulating insulin and IGF-1, thereby inhibiting tumor cell proliferation and enhancing the efficacy of cytotoxic chemotherapy. This specific application is being studied by Beni-Suef University in non-diabetic patients to improve progression-free survival.
