Overview

Metformin + nivolumab is a combination therapy that pairs the oral antidiabetic drug metformin with the immune checkpoint inhibitor nivolumab. This combination has been investigated for potential synergistic antitumor effects. Nivolumab is a programmed death-1 (PD-1) inhibitor that works by blocking the PD-1 receptor, enhancing T-cell immune responses against cancer cells. Metformin, traditionally used for diabetes management, has shown potential to elicit metabolic changes that favor decreased intratumoral oxygen consumption and may improve the efficacy of immunotherapy. The combination has been studied in various cancer types including microsatellite stable (MSS) metastatic colorectal cancer, non-small cell lung cancer, melanoma, renal cell carcinoma, and pancreatic cancer.
