metformin + ProMetS is a combination therapy consisting of the biguanide metformin and the probiotic formulation ProMetS. It is being investigated by The Affiliated Nanjing Drum Tower Hospital of Nanjing University Medical School for the treatment of polycystic ovary syndrome (PCOS) in reproductive-aged women. Metformin acts as an insulin sensitizer by activating AMP-activated protein kinase (AMPK) and reducing hepatic glucose production, thereby addressing the insulin resistance often associated with PCOS. ProMetS is a probiotic powder intended to modulate the gut microbiota composition, which is frequently dysbiotic in women with this condition. The combination aims to synergistically improve metabolic parameters, hormonal balance, and clinical symptoms such as menstrual irregularity and ovulation rates by targeting both systemic metabolism and the gut-brain-ovary axis.
