Drug intelligence / Profile preview
A fixed-dose combination of three oral antidiabetic medications: **metformin**, a biguanide that reduces hepatic glucose production and improves insulin sensitivity; an **SGLT2 inhibitor** (sodium-glucose cotransporter 2 inhibitor), which lowers blood sugar by blocking glucose reabsorption in the kidneys leading to glycosuria; and a **thiazolidinedione**, which improves insulin sensitivity via peroxisome proliferator-activated receptor gamma (PPARγ) agonism. This combination targets multiple pathogenic mechanisms in type 2 diabetes, aiming for enhanced glycemic control and potentially additive effects on cardiovascular and metabolic risk factors. This triple therapy is being studied for its efficacy, safety, and ability to minimize specific side effects common to each component, such as weight gain (from thiazolidinedione) and genitourinary infections (from SGLT2 inhibitors). Clinical evidence supports the complementary and potentially synergistic effects of this triple regimen on glycemic outcomes, as well as possible benefits in reducing fluid retention and weight gain associated with thiazolidinediones when combined with SGLT2 inhibitors[1][3].
