Overview

This is a combination therapy consisting of three distinct antidiabetic agents used in the management of type 2 diabetes mellitus: - **Metformin** (a biguanide) decreases hepatic glucose production, reduces intestinal absorption of glucose, and improves insulin sensitivity. - **Sulfonylurea** (class includes glipizide, glyburide, glimepiride) stimulates pancreatic beta cells to increase endogenous insulin secretion. - **Insulin** (exogenous hormone replacement) directly lowers blood glucose by facilitating cellular uptake and storage of glucose. This triple therapy is typically reserved for patients with type 2 diabetes who do not achieve adequate glycemic control with dual oral therapy. The combination leverages complementary mechanisms to improve blood sugar control but increases the risk for hypoglycemia and other adverse effects. There is no single fixed-dose product containing all three; rather, this regimen involves co-administration of individual drugs[1][4].
