Overview

metformin + turmeric is a combination therapy being evaluated in a Phase 1 feasibility study at the University of Arkansas for patients with biochemically progressive castration-sensitive prostate cancer (CSPC) receiving intermittent androgen deprivation therapy (iADT). The regimen consists of metformin, a biguanide small molecule traditionally used for type 2 diabetes, and turmeric, a nutraceutical containing the active compound curcumin. Metformin is thought to inhibit tumor growth by activating the AMP-activated protein kinase (AMPK) pathway, inhibiting mTOR, and reducing systemic insulin and IGF-1 levels. Turmeric/curcumin provides complementary activity by inhibiting androgen receptor (AR) signaling, downregulating NF-kB, and modulating various inflammatory and proliferative pathways. The study aims to determine if this nutritional and pharmacological intervention can safely delay PSA relapse in men with CSPC.
